CBC with Differential

Principle

The complete blood count (CBC) with 5-part differential is a comprehensive hematologic assay that quantifies circulating red blood cells, white blood cells, and platelets, along with several erythrocyte indices. The test includes measurement of red blood cell count (RBC), hemoglobin (Hgb), hematocrit (Hct), mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), red cell distribution width (RDW), platelet count, and white blood cell count (WBC). The 5-part differential provides both absolute and relative counts of neutrophils, lymphocytes, monocytes, eosinophils, and basophils.CBC) with 5-part differential is a comprehensive hematologic assay that quantifies circulating red blood cells, white blood cells, and platelets, along with several erythrocyte indices.

This assay is used to evaluate anemia, infection, inflammation, immune disorders, hematologic malignancies, and other conditions affecting blood cell production or survival.

Specimen Requirements

Specimen

Whole blood collected in EDTA (purple-top tube).

Collection

Mix gently for 2 minutes immediately after collection to prevent clotting.

Processing

Mix gently for 2 minutes immediately after collection to prevent clotting.

Storage and Transport

Keep and transport at room temperature.

Unacceptable Specimens

  • Specimens collected in tubes other than EDTA
  • Tubes less than half full
  • Improperly labeled specimens
  • Hemolyzed specimens
  • Specimens older than 48 hours
  • Clotted specimens
